I am building my very first rig from the ground up and was wondering if, BEFORE I install the water cooling "stuff", would testing my hardware and OS without any cooling for a very short period of time be safe?
I just don't want anything bad to happen...Very expensive rig (read my signature :p )

Yeah, I suppose the CPU would run for a half second or so before it overheated and shut down, not enough time for the OS to load though and the video card would surely fry.
Don't even think about doing that!
If you don't mind spending the the time, and since it's your first build, you may want to set everything up on air first. Then when all is well and good, install the WCing. That may save you alot of time in cast there are problems, as air cooled components are much easier to troubleshoot (i. e. remove and replace) than WC'd parts. That will also afford you the opportunity to record some temps for future comparisons.
It one thing to run for a moment without a fan on the heatsink or without the pump running (for like 3 seconds), but without a heatsink will cause near-instant hardwear death and possibly a fire

why not test his very expensive rig with stock cooling for a while...
(oh with intel stock HSF you usually dont have to apply the AS5)
but with the water block you do have to apply AS5....
