I just added a stick of 512 Mb of pc3200 400Mhz to my system.

My system,
Athlon xp2400 cpu
Gigabyte GA-7vaxp mb with promise onboard raid controller
Kingston value ram 1024 ddr pc3200 400mhz
Dual 80gig maxtor hd's 7200rpm ata 133 raid 0
Artec 16X DVD-ROM
Cyberdrive 40X16X48 CDRW
Verbatim 2.4X DVD+RW/R
ATI all in wonder 8500dv AGP primary video card
VisionTek XTASY radeon 9100 64mb ddr PCI 2nd video card
Nokia 21" 445Xpro CRT Monitor
KDS 19" visual sensations CRT monitor
Antec 430 watt true power ps
Ahanix platinum xp steel case
Windows xp home sp1

I've tried different things like recoding a dvd with nero and scanning a picture at 2400 resolution. The scanning accessed alot of the memory, but otherwise most of the additional memory remains unused. Are there any tweaks other than overclocking that will make my system use the extra memory.

Try reducing the size of the page file how ever dont it turn off as windows requires it to run. Basically if you reduce it you will be forcing it to use main memory (RAM)

try it and post back some results
