Is there confusion in what the manual says? If the manual or specs dont show it or have a block diagram, consider asking asus. Report back. :)

Id imagine it shares sata ports of some sort.

Edit: from the pro gaming review -
There are two M.2 SSD slots available, two 8cm type 2242/2260/2280. Both support PCIe x4/x2 Gen 3.0. The NVMe PCIe Gen3 x4 M.2 onboard connectors are however shared with the SATA connectors. So if you use the upper M.2 slot then SATA1 will be disabled. With the 2nd M.2 slot in use it means that port 5 and 6 are disabled.

Thanks it is the same as your quote for the other motherboard. I gave a call and it is in the manual page ix that I missed.
 
Indeed it is the same, but it was pulled from the ASUS Z370 Tuf Pro Gaming review while the other was sourced in his manual for his board.

Glad you found it. I assume it is the same as the Pro Gaming I quoted above?




EDIT: Looks like!

SATA1 will be disabled when using a SATA based device in M2_1.
SATA 5/6 will be disabled when any M.2 module is used in M2_2.
