yes both the NIC and mobo have to support WOL (make sure its also enabled in your BIOS) In order to use WOL you have to send a "magic packet" ping to the computer you wish to wake-up, when the NIC receives this signal, it sends a message to your ROM BIOS which, if enabled, will power on your computer and it will procede to boot up.

Some NICs also dont need a specific magic packet. I can set my card to wake-up the PC on any network traffic. So if my sister turns on her computer, it would turn on mine eventually.
 
Another thing that is necesary for wake up on lan is the cable that will conect the mobo to the NIC. it usaually comes with the NIC but not always.
