You'll need to make sure that "Shell Hardware Detection" is set to "Automatic" under "Services" (Start | Run | Type services.msc in the Run box. Right click "Shell Hardware Detection" and select "Properties" | "General" tab | Set "Startup Type" to "Automatic" (if it's set to anything other than "Automatic").

Right click the DVD drive in My Computer or Windows Explorer | Select "Properties" | "AutoPlay" tab....you should be able to enable/disable AutoPlay from here. You can also choose "an action to perform" from here, as far as which program should be used when opening a particular content type.

You can also use TweakUI to adjust the AutoPlay settings for your CD-ROM/DVD (instead of editing the Registry manually). In TweakUI under "My Computer" | Expand "AutoPlay", you can enable/disable AutoPlay for CD and DVD drives, enable/disable AutoPlay for removable drives, delete/create/edit AutoPlay handlers, etc.

You can download the TweakUI Powertoy from MS's site here...
